Background of the Study
CRISPR-Cas9 is a revolutionary gene-editing technology that has transformed molecular biology by enabling precise modifications to the DNA of living organisms. This tool has been widely used for genetic research, disease modeling, and therapeutic development (Shao et al., 2024). Despite its remarkable potential, the application of CRISPR in clinical settings requires accurate simulation tools to predict the effects of gene edits, minimize off-target effects, and enhance editing precision. Computational biology, which combines algorithms, data analysis, and simulation models, plays an essential role in optimizing CRISPR-based gene editing by simulating potential outcomes of gene modifications. Federal University, Birnin Kebbi, is exploring the use of computational biology to study the effects of CRISPR gene edits on specific diseases. This study aims to develop and apply simulation models to predict the outcomes of CRISPR gene editing, focusing on optimizing gene-editing strategies and minimizing associated risks.

Statement of the Problem
CRISPR-based gene editing holds great promise for treating genetic diseases, but the unpredictability of off-target effects and incomplete edits remain significant concerns. While experimental techniques provide valuable insights into CRISPR’s applications, computational biology can aid in predicting the likely outcomes of gene modifications and in optimizing editing strategies (Zhao et al., 2025). At Federal University, Birnin Kebbi, computational biology tools are yet to be fully integrated into CRISPR-based research, limiting the ability to optimize gene editing techniques for specific disease targets. The study will address these issues by simulating the effects of CRISPR gene editing on disease-related genes, aiming to enhance the precision and safety of gene-editing interventions.

Objectives of the Study

  1. To develop computational models for simulating CRISPR-based gene editing on specific target genes.

  2. To analyze the efficiency and accuracy of CRISPR edits through computational simulations.

  3. To identify potential off-target effects and strategies to minimize them using computational biology techniques.

Research Questions

  1. How can computational simulations improve the accuracy and efficiency of CRISPR-based gene editing?

  2. What are the potential off-target effects of CRISPR edits, and how can they be minimized using computational models?

  3. How effective are the computational models in predicting the outcomes of CRISPR gene edits in disease-related genes?

Significance of the Study
This study will enhance the application of CRISPR-Cas9 gene editing by improving the precision and safety of gene modifications through computational biology simulations. By optimizing gene-editing strategies, the research will contribute to the development of more effective and personalized treatments for genetic diseases, with a particular focus on those prevalent in Nigerian populations.

Scope and Limitations of the Study
The study will focus on developing and implementing computational models for simulating CRISPR-based gene editing targeting disease-related genes. It will be conducted at Federal University, Birnin Kebbi, Kebbi State. The limitations of the study include the reliance on available genomic data and the computational resources required to run large-scale simulations.
